Packaging of DNA into bacteriophage heads: a model.
نویسنده
چکیده
A model is suggested for the geometry of DNA entry into a bacteriophage head. It accounts for recent observations indicating absence of a unique, ordered sequence of windings in the packaged DNA.

A Promiscuous DNA Packaging Machine from Bacteriophage T4
Complex viruses are assembled from simple protein subunits by sequential and irreversible assembly. During genome packaging in bacteriophages, a powerful molecular motor assembles at the special portal vertex of an empty prohead to initiate packaging. The capsid expands after about 10%-25% of the genome is packaged. When the head is full, the motor cuts the concatemeric DNA and dissociates from...

Bacteriophage DNA packaging motors translocate their genomic DNA into viral heads, compacting it to near-crystalline density. The Bacillus subtilis phage 29 has a unique ring of RNA (pRNA) that is an essential component of its motor, serving as a scaffold for the packaging ATPase. Previously, deletion of a three-base bulge (18-CCA-20) in the pRNA A-helix was shown to abolish packaging activity....
متن کاملPackaging recombinant DNA molecules into bacteriophage particles in vitro.
Recombinant phage genomes made in reactions with purified enzymes may be recovered directly by packaging into phage heads in vitro. The process is efficient and nonselective and offers containment in initial stages of handling recombinant DNA. Ligase [poly(deoxyribonucleotide):poly-(deoxyribonucleotide) ligase (AMP-forming), EC 6.5.1.1] reaction products can recombine with endogenous phage DNA ...
متن کاملSymmetry mismatch and DNA packaging in large bacteriophages.
A model is presented for the mechanism of packaging double-stranded DNA into phage heads. The model is based on, and rationalizes, the mismatch in symmetry between the heads and tails of large bacteriophages. DNA movement is postulated to be mediated by a rotating protein structure at the tail-proximal vertex of the head.
متن کاملStructure-function analysis of the DNA translocating portal of the bacteriophage T4 packaging machine.
Tailed bacteriophages and herpesviruses consist of a structurally well conserved dodecameric portal at a special 5-fold vertex of the capsid. The portal plays critical roles in head assembly, genome packaging, neck/tail attachment, and genome ejection. Although the structures of portals from phages φ29, SPP1, and P22 have been determined, their mechanistic roles have not been well understood. S...
